I applied online and got called for an interview. I came in, waited for the manager and did my interview. I was asked about 5-8 questions. He told me he would call me if he was interested. Later that day I was called back and ask if I was okay with a background check. I agreed and got a call back in about a week for orientation.

I applied online, got a call 3 days later. They setup an interview for the following day. I had to dress formally.
They asked maybe 10 questions. The questions were picked at random from a packet of questions. The packet contained around 3 questions per page, and they selected 1 one them, then flipped the page.
Some questions I remember:
-What do you know about Duane Reade?
-Give me a scenario where you went out of your way to help a customer.
-Why do you want to work at Duane Reade?
-What do you think it means to keep the store clean/neat?
-Give me a scenario where you had to deal with a bad customer?
-What do you think good customer service means?

Applied online and was contacted by a manager a few days later. Went in for a one on one interview which was a few basic questions about previous experience (don't worry if you don't have any),customer service and why Duane Reade, etc. Manager was friendly and the interview was relaxed and conversational. Received the offer a few days later.
